Case Study: Development of a Scalable LV Production Platform

Developing lentiviral vector production processes that scale reliably across different bioreactor platforms requires more than increasing volume. Cell line behaviour, transfection conditions and engineering parameters all interact and can influence vector yield, consistency and process robustness.

What you’ll learn
In this webinar, Lindsay Mulder, Scientist Viral Vector, will present a case study on upstream development of a suspension-based lentiviral vector production platform using HEK293T cells.
The session builds on the concepts introduced in Lodewijk’s previous webinar and provides a step-by-step view of how the process was established and scaled.
Lindsay will discuss a practical case study, where adherent HEK293T cells were adapted to suspension culture and how the resulting cell line was established and characterised for stable GMP vector production.
This webinar will address key questions, including:
  • How does lentiviral vector production differ between adherent and suspension systems?
  • What are the key steps in creating a robust suspension cell bank?
  • How can cell line suitability for GMP production be assessed?
  • What strategies can be used to optimise transfection efficiency?
  • How can scale-up in stirred tank reactors (STRs) be effectively optimised?
  • What considerations are important when designing a scale-up pathway?
  • How can processes be successfully transferred into a GMP environment?
